Touchdown pcr6/23/2023 Temperature profile for Touchdown PCRĪny difference in the Tm between the correct and incorrect annealing temperatures will result in an exponential increase by 2-fold per cycle. Reaction cycle for Touchdown PCRįor example, in a PCR, the Tm temperature for a pair of primers is 55º C, then in touchdown PCR, we will set the first annealing temperature at 65º C and then gradually decrease by a degree Celsius. The second phase is generic PCR amplification with final annealing temperature of 20-25 cycles. Thus, after each cycle, the temperature is decreased by 1º C, till it reaches the desired range of Tm, this is first phase of TD-PCR consisting of 10-15 cycles. For this we select the annealing temperature 5 º C to 10º C higher than the Tm value, so that the primers can bind only to its specific and complementary sequence at appropriate temperature. This gradual lowering of temperature, so that the annealing temperatures can be more suitable during the course of cycling favors amplification of the desired amplicon. Touchdown PCR is a modification of PCR in which the initial annealing temperature is higher than the optimal Tm of primers and then it is gradually reduced over subsequent cycles until the Tm temperatures are reached for the primers. In order to avoid this, there is simple and rapid means to optimize PCR specificity, sensitivity and yield without these lengthy optimizations and primer redesigning, i.e., the touchdown PCR. But primer designing is a lengthy optimization and designing process. When the primer annealing is a bit difficult, i.e., there is temperature difference between the melting temperatures (Tm) of forward and reverse primers, there is need of redesigning the primers. Touchdown PCR is for increasing the amplification and specificity of PCR reactions. ![]() One such variant is Touchdown PCR (TD-PCR). Many variants of the technique have been developed over the course of time. Flooding in webber falls ok6/23/2023 For telling me when I look beautiful, and being honest with me when I don't. For giving me your nose, and your spirit. For telling me how to do better, and recognizing the pride I have when I give it my all. The truth is I could never thank you enough. Thankfully, you taught me how to be just like you. How to be feminine and strong all at once. You taught me how to live like a Proverbs 31 woman. But at the same time I am more than ready. It'll be a whole new world for the both of us. So how do we move away from these feelings of self doubt and self hate as a society? How do we better inform viewers and consumers that all body types are acceptable? I'll be back with more on this next week. Thus, the vicious cycle of never feeling good enough is born. Failed diets and failed exercise routines lead us to feel even worse than when we started. The exercise world can, in turn, be no better certain workout plans believe that cardio, and lots of it, are the only way women will get thin, or the plans start in an advanced stage and the customer, in turn, cannot keep us as they are beginners, so the plan is ultimately dropped. With crazy diets telling us to do things like eating half of the calories we burn every day, only drinking water, lemon, and maple syrup for 7 days as all our meals, only eating when you feel like you're going to pass out, and so on and so forth, the health of women and the sustainability of these ridiculous diets start to become a real concern. With the media's power over our gaze, lots of women then turn to diet and exercise to try and fit into society's ideal figure. When the average representation of what the perfect figure for women looks like turns out to be an incredibly thin, curve free model, it's hard for women who are not born with this body type to accept that how they look is more than okay. Children today often look up to other child actors, or what is portrayed as a child actor, and think, "why doesn't my body look like that? Aren't we the same age?" Growing up is hard enough, but growing up with the image of an 18 year old playing the age of a 14 year old on tv is incredibly difficult for viewers to see and it doesn't stop there as we get older, we look to magazines, fashion instagrams, influencers, and other media outlets to see what the latest trends are, but most importantly, we look to see what we should look like. Diets, exercise crazes, and the media's portrayal of the 'ideal body' has been a major contributor to women and young girls, as well as men, believing their natural body shape is not acceptable. How did a time once filled with nothing but joy turn into this doom of "will my body be good enough"? America has grown increasingly more hostile towards body image over the past 50 years. Over the past several weeks, I've gotten many nervous "7 weeks until bikini time," "4 weeks until bikini season," "2 weeks until I have to be in a bikini," "I'm not going to the beach or any pool parties this summer" texts from several different friends. Main stream media has begun to include all body types in modeling and advertisement campaigns, but yet there's a sense of not feeling good enough still lingering in the air. Summer is the warmest and most fun season, but it can also easily be the most stressful season for anyone who suffers from body image issues. Whenever I give a tour of my town I never leave out the village or the colleges because they’re such a huge part of what makes my town so special. In fact, Claremont is known as a college town because of how college centered it is (so much so we actually named a lot of our streets after famous colleges). They all have their own personalities despite not having any distinct borders separating them. They’re all very distinguished, both in their reputation as educational institutions and their campus. They’re known as the Five-C’s (The Five Claremont Colleges): Pomona, McKenna, Scripps, Pitzer, and Harvey Mudd. What also contributes heavily to the grandeur of the city is the fact that it’s the home of five very prestigious colleges. ![]() You’ll rarely find the restaurants and shopping attractions anywhere else. It has a village that’s filled with unique, privately managed businesses that further add to the uniqueness of the overall city. Nevertheless its beauties are unbound by the restrictions of descriptive words as they do it no justice. Claremont is about an hour or so away from L.A. Blood island the day may come soon lorca6/23/2023 ![]() It's just too funny when Lorca is addressing the severed head, and even more so when the head is seen to speak! A pretty good finish and highly amusing gore and creature effects add to the appeal of this movie which if not great is still pretty good. Familiar faces Alfonso Carvajal as tribe leader Ramu and Bruno Punzalan as henchman Razak are fun, but not as much as Garcia, who seems to be relishing his role Lorca cheerfully discloses at one point that he's "madder than ever". Yarnall, known for appearances in movies like "Live a Little, Love a Little", "The Velvet Vampire", and "The Mechanic", is super sexy and a commendably, fairly feisty type, and Liza Belmonte is also a real looker as helpful local Laida. The acting basically gets the job done, with Ashley effective as the stolid hero. But, as with any of these Filipino horror movies, the inherent atmosphere of the locale comes through strong, and accompanied by the expectedly (and endearingly) grandiose music by Tito Arevalo, "Beast of Blood" does have the perfect "midnight movie" feel to it, and does have the legendary Eddie Romero in its director's chair if you watch enough of these movies, Romero's is one name you'll start to see repeatedly. Lorca is keeping the severed head of the monster alive in his lab, for one thing! This is overall not as much fun as one might wish it to be, as it has a mostly uneventful first half, and the movie does mostly consist of a trek / pursuit through the jungle. Lorca, now played by Filipino film veteran Eddie Garcia, is very much alive and still up to no good. He's now in the company of nosey, stubborn reporter Myra Russell (Celeste Yarnall) and a captain played by Beverly Miller (Miller also concocted the story for this entry). Bill is rescued, and one year later he returns to the island upon hearing stories of what may still be going on there, even though the villainous Dr. Bill Foster (John Ashley) is sailing away from the island when the monster of "Mad Doctor" causes the destruction of the boat. "Beast of Blood" picks up where its predecessor, "Mad Doctor of Blood Island", leaves off, so in essence both movies are one long story. Definitely worth seeing especially as part of a double feature with Mad Doctor of Blood Island. What can I say, I took it on its own terms and I liked it, high art its not. Its not bad, but if you are expecting a straight horror film you may end up very disappointed, despite a great monster. Some of Lorca's victims do cause mayhem, but the majority of the film concerns trying to find Lorca and the kidnapped reporter. The man-beast is effectively off camera for most of the film following the opening battle (I have to say the make up here is infinitely better than in Mad Doctor). Sure there are some horrifying moments, but on some level this is more an adventure/ mystery film than a real horror movie. A sequel it is, but its not as scary (nor as gory nor as titillating). What it was missing was the set up that the first film gives it.If you watch the two films together I think you'll find it a better film than when it's viewed all alone. I was never a big fan of this film prior to the back to back viewing because I always felt that it was missing something. Finally watching this in close proximity to Mad Doctor I found that the film plays much better than it does as a stand alone film. It isn't long before its realized that Dr Lorca is back and up to his old tricks. On the island Ashley finds that many people he believed dead survived the final battle of the first film and that some how the "green men" have returned. A year or so later Ashley heads back to Blood Island to investigate stories that weird things have begun to happen again despite the death of the evil Dr Lorca. ![]() As John Ashley waxes poetic about his time on the island fighting monsters the man beast from the first film appears (he was seen to have secreted himself in a lifeboat at the end of Mad Doctor) and a battle occurs which destroys the ship and leaves Ashley as the only survivor. Literally picking up hours after the first film, the film begins on the boat sailing away from Blood Island. Its the fourth (or third if you don't count the unrelated Terror is a Man) in the Blood Island saga. This the direct sequel to Mad Doctor of Blood Island. Hints of alan wake 26/22/2023 ![]() ![]() By tactically fleeing to the next 'Safe Haven' checkpoint, you will be actively conserving your scarce supplies for the fights that you have to take. Alan Wake is a Survival game, and often the key to surviving is getting the hell out of there. Alan Wake isn't meant played as a First Person Shooter you do not need to kill every enemy all the time. The only important suggestion in this game is: learn how to escape. In the Walkthrough, everything which is calculated in the stats will be described the rest might or might not be mentioned, since it's not necessary (and doesn't add anything to the gameplay anyway). There are more than 25 Signs, and the extra ones are not related to any achievement/won't count for the stats (same as for the radio shows). There are more than 11 Radio Shows (actually, the extra ones are not radio shows but random transmissions), but the extra ones are not related to any achievement, and they won't count for the stats neither (this means you can't collect one of the "extra ones" instead of one of the regular shows, because it wouldn't count). In particular, there are 12 Can Pyramids and you only need 5 for the achievement related to them. With the short loading times of this game, this practically means having infinite ammo and infinite enemies to collect kills and mop up those achievements in a short time. ![]() The above note is also true for any other statistic, and this makes the "restart checkpoint" a valuable help for grinding kills with your weapons, since your stats will be updated if you restart a checkpoint, and you also get your ammo back (provided you had them when you hit the last checkpoint which you will restart). In other words, you don't have to worry about collecting the same item again if you die/restart a checkpoint, because as long as it was saved in your statistics it will count for the achievement. If you die after collecting something, and restart a checkpoint triggered before picking it up, that item will be in its specific location again, but you won't need to collect it again for achievement purposes since the statistics were already updated (and they are what matter for the achievements). You will clearly see the saving icon in the bottom-left corner when you collect something. Collectibles will be saved to your statistics (you can check them pressing while playing) as soon as you pick them up for the first time. Hearts of iron sabaton6/22/2023 It was great fun to record that solo as well because Thobbe, our guitar player, was supposed to do it and he sometimes gets nervous in the studio when you press record. Accept did “Für Elise” on the Metal heart song and we thought it was brilliant, and finally when we had the song “Hearts of Iron” we were listening to the chords of it and wow, this actually has the same chords as “Air on a G string”, so we can put that in here. We are inspired by Accept, there’s no need to say anything else. Pär: "Yes it is, it’s “Air on a G string”. "In Hearts of Iron, is that a piece of classical music thats played as a guitar solo? It sounds very familiar but I cant quite place it." It's pretty funny actually today, but a lot of our fans in China discovered Sabaton through the game Hearts of Iron." So it was possible to get the music, our songs, when you play the game. note), there is release now soundtrack of Sabaton. We did other things connected to this, and inside Hearts of Iron (IV - ed. And when we were researching we came across the story about Walter Wenck and it took a while to keep discussing and developing the idea. They asked us "is there any way that you can like write a song that would be like a lead theme for our game that is coming?" and we said "okay". I mean they are doing games about history and we are doing music about it. Pär: "Many years ago, when we had a concert in Stockholm, we were approached at an after party by people from this Swedish development studio of games called Paradox, and some of the guys came and they said that they were big fans of us and they really appreciate the work that we are doing with a music about history and we started to talk about that we are doing pretty much the same thing. In fact, he pressed "record" during the practice and the take was perfect so it ended up on the album. The guitar solo was recored in one take by Thobbe Englund, who was tricked by the producer Peter Tägtgren, who said that he's going to the bathroom and they will record when he gets back, so in the meantime Thobbe can practice the solo.It was inspired by Accept's song "Metal Heart" that used Beethoven's "Für Elise" as the guitar solo. The guitar solo in the song is Bach's "Air on the G String".Also, in German instead of "Not a battle" which the band used, "No battle" ("Keine Schlacht") would be the correct form. It should be " Keine Schlacht, eine Rettungsaktion" since both Schlacht and Rettungsaktion are of female genus in German. ![]() The song topic comes from the book "Endgame" by David Stafford which Joakim got from a fan.The band was asked to make theme song for the Hearts of Iron game, but some time later they came across the story of Walter Wenck and it took a while to actually compose the right song for the topic. The song title was inspired by a video game series "Hearts of Iron". The house that jack built6/22/2023 ![]() Massumi explores the function of art in Marcel Proust’s diagrammatic work in In Search of Lost Time, where the description of a madeleine cookie and lime tea leads to other diagram-icons in the text such as the grandmother’s kiss and the wart on her cheek. By this I mean such a transformation of our diagrams that characters of one diagram may appear in another as things. The greatest point of art consists in the introduction of suitable abstractions. ![]() I was inspired by Brian Massumi’s emphasis on Peirce’s reading of the task of art as related to the art of creating diagrammatic abstractions. ![]() What has guided my non-representative reading is the immense affective impact his films have had on audiences everywhere. In that book, I studied von Trier’s oeuvre of film in relation to the exploration of the material qualities of pixels and signals in film and video as well as the diagrammatic style used extensively from the mid-1990s. This article, then, is a further development in an argument that I began in my recent book Lars von Trier’s Renewal of Film 1984-2014: Signal, Pixel, Diagram. They can provide ‘a viewpoint or standard according to which phenomena are seen and determined’. They do not simply depict the (already) visible they make visible the (hitherto) invisible. Such diagram-iconsĪre defined in dynamic and operational terms. I suggest von Trier’s emphatic use of iconicity in the film creates what I will mostly refer to as diagrams or ‘diagram-icons’ that articulate, as already indicated, a critical assessment of iconocity itself. My argument departs from Peirce’s idea that icons can also be diagrams and establish analogue (often dyadic) relations to (parts of) an object. The icon – covering three types: image, diagram, metaphor – shares qualities with the object it resembles or imitates. Following Peirce, the indexical sign carries a relationship with an object (a photographic print as an index of light passing through a camera lens) whereas the symbolic sign is a unifying, often arbitrary sign referring to cultural convention (a cross standing for Christianity). ![]() In the following reading, I will make use of Charles Sanders Peirce’s semiotic definitions of icon, index, and symbol. Photographic and filmic representations have often been referred to as indexical signs, but as images they are also iconic signs. It is at this level that the film articulates its assessment of Western image regimes as emphatically iconic, a quality which provides for both their aesthetic promise and their political danger. In this article, I intend to show that the film’s transversal relations between iconic features in the diegesis and various explorations of what characterises icons on a cultural level pave the way for abstractions. Thus, Jack’s useless efforts to create an iconic masterpiece of architecture while murdering forms the diegetic world commented on from Verge’s initially extradiegetic voice, later integrated as a character. Meanwhile, the film’s real forger is Verge (Bruno Ganz), a figure who first appears as voiceover commentary who mocks and undoes Jack’s ambitions – both as a murderer and an architect/artist. But even though he is a criminal, his ambition is another: to become an architect and build an iconic house. The main character, Jack (Matt Dillon), is an engineer and mass murderer, who challenges the laws of society and gets away with it. Lars von Trier’s most recent film, The House That Jack Built, premiered in 2018. |
